The Greek term "Pharmakon" was used for poison, at the same time for antidote and medicine. During centuries, poisonous and healing plants have been detected by trial and error; it was not before 1800, however, that natural science became successful in isolating and identifying active substances from plants. With the development of chemistry the number of known structures increased enormously, and in many cases their mode of action became explainable. Albert Hofmann in his work, sometimes led by serendipity, gives us excellent examples for classical natural products chemistry. To which extent is it possible to find and improve natural compounds for their use in medicine? Rudolf Brenneisen Cannabis - From Phytocannabinoids to Endocannabinoids The Cannabis plant has been an essential element of traditional medicine for thousands of years. Today, its medicinal use is becoming again popular mainly within self-medication. However, the discrepancy between empirical and evidence-based data is obvious and therefore implies intensive pharmacological and clinical research. On the other hand, the recently discovered Cannabis receptors and their endogenous ligands are potential targets for new therapeutic tools. Matthias Hamburger Contemporary Natural Product Drug Discovery Natural products have been the historically most prolific source for drugs and inspiration for designing synthetic drug substances. Recently, their role in drug discovery has been challenged by the advent of combinatorial synthesis and high-throughput screening. Contemporary opportunities for natural products will be discussed in the larger context of methodological advances and new paradigms in the life sciences. Selected examples will highlight the continued importance of natural products in target discovery and as source for new drug templates with unique properties. Why I stopped using products with parabens...with research

PLEASE PLEASE READ!!!!: *Disclaimer*…putting this right up front. Today’s video is my personal experiences with makeup, my choice to give up products with parabens, and my interpretation of current research on the use of parabens in cosmetics. This video is in no way a judgement against companies that continue to use parabens or consumers who purchase those products. As I explain in the video, I use products with other chemical ingredients, so I don’t claim to be a “green beauty” consumer, though I do use a blend of all natural products and less natural products that just don't have parabens in them. Please look below for the articles that I reference in the video. Cosmetic company I mentioned: Larenim Minerals - http://larenim.com/ Lots of great products, and you can get sample sizes for only $4. Because a lot of their stuff is so highly pigmented I find that the samples can last me a really long time. So, in preparation for this video I looked at a number of articles, most of them from toxicology journals, to look at what the current scientific research says about the use of parabens in cosmetics. Here is a sampling of what I found: *Braun, Joe M., et al. “Personal care product use and urinary phthalate metabolite and paraben concentrations during pregnancy among women from a fertility clinic.” Journal of Exposure Science and Environmental Epidemiology. 24 (2014): 459-466.* “Our previous work in this cohort did not suggest that pregnancy-induced changes in this paraben or phthalate metabolite concentrations before or during pregnancy. However, unwary phthalate metabolite and paraben concentrations have moderate within-person variability during pregnancy due in part to eh short biological half-life of these compounds. This may necessitate multiple spot urine samples to classify gestational exposure. Indeed, a strength of the current study was the repeated urine measurements, allowing us to incorporate this variability into our estimates.” (465) *Hu, Pan, et al. “Effects of Parabens on Adipocyte Differentiation.” Toxicological Sciences. 13.1 (2013): 56-70.* “Although future studies are required to delineate the molecular mechanisms by which parabens act on adipogenesis in vitro and to define the parabens’ action in vivo, our results may have uncovered a potential link between the increasing prevalence of obesity and the constant environmental exposure to parabens.” (68) *van Meeuwen, J.A.. “Aromatase inhibiting and combined estrogenic effects of parabens and estrogenic effects of other additives in cosmetics.” Toxicology and Applied Pharmacology 230 (2008): 372-382.* “In conclusion, it is obvious that actual concentrations found in human samples for single parabens (in the range of 10-80 nM) are 2 to 4 orders of magnitude lower than effective concentrations in in vivo systems such as cell proliferation and aromatase inhibition in placental microsomes. However, the list of chemicals exerting potential endocrine active effects grows as does the number of consumer products available containing these chemicals. On top of that, for such chemicals the evidence for an additive effect on endocrine sensitive endpoints accumulates.” (380) *Boberg, Julie, et al. “Possible endocrine disrupting effects of parabens and their metabolites.” Reproductive Toxicology. 30 (2010): 301-312.* This was probably my favorite article. There was so much good information in here there’s no way I can quote all of it. This was the article that talked about acceptable daily intakes, though as I mention in the video, it’s hard to make sense of the number they provide since there’s no way to calculate what amount of parabens we take in on a daily basis since levels are not posted on cosmetic labels. This is also the article that discusses the estrogenic burden of parabens in children. A lot of this is still hypothesis and longer term studies are needed but they suggest, “Estrogenic effects in boys may increase the risk for incomplete masculinization resulting in decreased sperm quality. In girls, an increased estrogenic load may increase the risk of early puberty, premature mammary development and the risk of mammary cancer.” (310) *Harvey, Philip W. “Parabens, Oestrogenicity, Underarm Cosmetics and Breast Cancer: a Perspective on a Hypothesis.” Journal of Applied Toxicology. 23 (2003): 285-288.* This is an editorial on the recent studies at the time. The other studies I read had much more updated information. But this was a good starting point. Disclaimer: Again, this is just my perspective. The Takiwasi Center has just renewed its Scientific Research Department, which is focused primarily on the two main work areas of the Center: the Clinical Area and the Medicinal Plants Area in collaboration with the Research & Development Unit of Takiwasi’ s Natural Products Laboratory. The clinical protocol of Takiwasi is based on the combination of psychotherapeutic techniques with tools derived from Traditional Amazonian Medicine, including the ritual use of medicinal plants. The Natural Products Laboratory develops with commercial purpose herbal medicines based on local ancestral knowledge. Both activities have been developed over time, allocating little resources to the scientific dissemination of the results, despite the high value of the same and the great interest on our work on the part of international researchers. The renewed Scientific Research Department aims to strengthen research on the subject of traditional and herbal medicine, and its main objectives are: the publication of papers in indexed journals, the participation in scientific congresses and the application to research grant opportunities. These ambitious goals entail investments to be able to bring together specialized personnel in various fields including anthropology, botany, pharmaceutical chemistry, medicine, psychiatry and psychotherapy, and provide them with the necessary resources to implement the various research topics. Plants and microbes produce a wide spectrum of natural products, which give them their huge range of colours, flavours and scents. These chemicals are used to repel pests, diseases or competitors or to attract pollinators, but for humans they represent a potential source of new medicines, flavourings, antimicrobials or other useful compounds. A major area of study at the John Innes Centre is in understanding how and why plants and microbes make these natural products. With this knowledge, we are looking at ways of exploiting them to help maintain our health, to improve crops plants and to maintain a safe, nutritious food supply. Professor Anne Osbourn leads a research group investigating the structure, function and evolution of natural products, in particular in crop and model plants. Professor Cathie Martin's research background is in cellular specialisation; in particular how flower colour and shape is determined. More recently she has been investigating how plants can be improved by manipulating the natural products that they produce.

It was mixed with crushed terracotta as an aggregate and formed into a cube. The cement set hard after 3 days and did not dissolve in water after this period. Process: First I burnt bark and leaves in a kiln at high temperatures to produce well burnt, mostly white wood ash. The ash was then mixed into water and stirred well. The excess water was poured off and the resulting paste was made into pellets and allowed to dry. A pellet was then re-heated in the forge until it glowed about orange hot. This was then taken out, cooled and dropped in a pot of water. The pellet dissolved and boiled due to a chemical reaction with the water. The paste was stirred and crushed terracotta (old tiles from previous projects) was added and mixed to form a mouldable mortar. This was formed into a cube and allowed to set for three days (in the video, a cube made exactly the same way 3 days previously was used due to time constraints). The resultant cube was strong and made a slight ringing sound when tapped with a finger nail. It was placed in water for 24 hours to simulate a very heavy rain event and did not dissolve or release residues into the water. My current theory: The main component of wood ash consists of calcium in some form (e.g. calcium carbonate, calcium oxide). This can be up to 45% from my research. Calcium is in higher concentration in the bark and leaves of a tree. When the ash is mixed with water, the soluble component of wood ash (10% pot ash) dissolves into the water. But seeing that it does nothing for the cementing process, it is drained off leaving the insoluble calcium (and other components) in the paste. Doing this probably raises the relative percentage of calcium in the paste to about 50% or more. Most of the other 50 % consists of silica and alumina which are pozzolans, materials that chemically react with calcium hydroxide to increase the durability of the cement product. The paste was then made into a pellet and fired again to high temperature to convert all the calcium compounds to calcium oxide. It also reduces any charcoal in the pellet to ash if it hadn’t already been burnt the first time. This step seemed important as un-fired ash pellets only partially hardened and would fall apart in water, though retaining a weak undissolved 5mm thick crust. I can only surmise that re-firing the ash just gave a greater conversion of the calcium components to calcium oxide. The pellet is slaked in water converting the calcium oxide to calcium hydroxide. This cement was mixed with crushed terracotta which may also help in some way that I’m not aware of as I only did this one experiment and did not test other aggregates yet (e.g. sand, gravel etc.). Terracotta is porous and might hold together better than other materials. The mixture is allowed to set in air where carbon dioxide reacts with calcium hydroxide to form calcium carbonate cementing the aggregate together. After this, the cement will not dissolve in water. Use: I think this material might have a potential use as a mortar holding rocks or bricks together in wet environments where limestone or snail shells are unavailable for making cement. Wood ash is a pretty ubiquitous material to most natural environments inhabited by people using biomass fuels. Wood ash cement turns a waste product into a valuable building material. From my research, wood ash is already being used as a partial replacement for cement in the building industry without decreases in strength of the final product. But I’ve only just started experimenting with it and don’t know its full capabilities and limitations. Calcium content of wood ash differs depending on the species of tree, the part of the tree burnt and the soil it’s grown on. Cautious experimentation is still required before committing to a hut built from this material.
